DNA collected from artificial shelter surfaces detected reptiles that visual checks missed.

Sampling the shelter after the animal left

A 2022 study added environmental-DNA sampling to a Wharton State Forest reptile survey. Researchers rolled sampling tools over metal and wooden cover objects and tested for little brown skink DNA. The comparison asked whether traces on a shelter could reveal use even when lifting it exposed no visible animal. Kyle and colleagues, reptile detection (2022).

More detections, with an important sampling qualification

Across 284 surface samples, the roller method detected skink DNA in 64 percent, compared with visual detections in 11 percent. The team deliberately sampled shelters associated with recent sightings and their paired objects; this was not a random census of the entire forest. The percentages therefore compare methods within the sampling design. Kyle and colleagues, reptile detection (2022).

The surface mattered

Metal produced more positive samples than wood. Soil sampling also improved detection when enough material was collected. The authors argued for combining approaches, while acknowledging uncertainty about deposition, persistence and movement of DNA. A positive trace identifies evidence of use, not the number of skinks present or an animal waiting underneath at that moment. Kyle and colleagues, reptile detection (2022).

Sources and research limits

Surface DNA and visual observations have different time windows. Raw assay files were not reanalyzed.
